المستخلص : In this study the effect of ranitidine, a well known H-2 receptor antagonist, on duodenal motility was investigated. 28 male albino rats were classified into 3 groups, 12 rats used as a control group, 8 rats injected with single dose of ranitidine and a third group of 8 rats received daily IP injection of ranitidine for one week. In vitro effects of ranitidine were also recorded. The response to ranitidine infusion in-vitro showed significant increase in both average force of duodenal contraction and the motlity index compared to their basal values. On the other hand the frequency of duodenal contractions and the average duration were not significantly changed with ranitidine infusion compared to the basal values. The addition of ranitidine after atropinization was still associated with significant increase in average force of contraction and motility index as compared to basal values. Although the degree of significance has decreased after addition of atropine compared with that before atropinization. In addition, ranitidine infusion augments the stimulatory effects of histamine on duodenal motility suggesting that ranitidlne has some histamine receptor agonistic action. In-vivo study showed also that injection of ranitidine as a single dose was associated with significant increase in the average force of contraction of the duodenum of injected rats compared to control group. Also, there was significant increase in motility index of single dose ranitidine injected rats. On the other hand, the freqwency of contractions and the average duration of contraction were not significantly altered. Similarly, rats daily injected with ranitidine for one week exhibited a similar motility pattern with higher degree of significance It could be concluded that, the H2 receptor antagonist (ranitidine) has a clear contractile effect on rat duodenum an effect that may be attributed to direct effect, a partial cholinergic effect and may be related to gastric acid suppression.
